How to Reconstitute Peptides: A Step-by-Step Lab Guide

Lab Techniques — 2026-04-08

Proper reconstitution of lyophilized (freeze-dried) peptides is a fundamental laboratory skill that directly impacts research quality and reproducibility. This guide covers best practices for peptide preparation.

What You'll Need

Step-by-Step Reconstitution

Step 1: Preparation Allow the peptide vial and bacteriostatic water to reach room temperature. Clean your workspace and wash your hands. Gather all materials.

Step 2: Clean the Vial Tops Swab the tops of both the peptide vial and the bacteriostatic water vial with alcohol pads. Allow to air dry.

Step 3: Draw the Bacteriostatic Water Using a sterile syringe, draw the appropriate volume of bacteriostatic water. The amount depends on your desired concentration — common dilutions are 1-2 mL per vial.

Step 4: Add Water to the Peptide Insert the needle into the peptide vial at an angle, directing the water stream against the inside wall of the vial — NOT directly onto the lyophilized powder. This prevents damaging the peptide structure.

Step 5: Allow to Dissolve Let the water gently flow down the vial wall onto the powder. Do NOT shake the vial. Instead, gently swirl or roll the vial between your palms until the powder is fully dissolved. This may take 1-5 minutes.

Step 6: Verify Complete Dissolution The solution should be clear and free of visible particles. If cloudiness persists, allow more time for dissolution or gently swirl again.

Why Bacteriostatic Water?

Bacteriostatic water contains 0.9% benzyl alcohol, which prevents microbial growth. This allows the reconstituted peptide to be used multiple times over up to 28 days when stored properly. Plain sterile water should only be used for single-use applications.

Storage After Reconstitution

Common Mistakes to Avoid

  1. Spraying water directly on the powder — This can denature the peptide
  2. Shaking the vial — Aggressive agitation can damage peptide bonds
  3. Using the wrong diluent — Always use bacteriostatic water for multi-use
  4. Improper storage — Room temperature storage dramatically reduces peptide stability
