Dosing — 2026-05-08
Most researchers new to reta don't fail because they pick the wrong target dose — they fail because they jump to a target dose too fast and discontinue from GI side effects in week 2. The Phase 2 trial protocol starts at 2 mg weekly, but in practice many research programs use a gentler micro-titration into the 2 mg starting dose to give the gut more adaptation time. This article walks through the practical "20 / 30 / 40 unit" beginner protocol, why it works, and where it sits relative to the published trial schedule.
Quick note for researchers sourcing material: code WELCOME at checkout unlocks 35% off reta 10mg, 20mg, bacteriostatic water, and the Metabolic Stack. Every vial includes a batch-matched COA — variable potency between batches is the single biggest confounder in beginner titration studies.
For a reta 10mg vial reconstituted with 2 mL of bacteriostatic water (yielding 5 mg/mL), this is the beginner ramp:
| Week | Dose (units on U-100 syringe) | Volume | Actual mg |
| Week 1 | 20 units | 0.20 mL | 1.0 mg |
| Week 2 | 30 units | 0.30 mL | 1.5 mg |
| Week 3 onward | 40 units | 0.40 mL | 2.0 mg |
Once you're stable at 40 units (2.0 mg) for 3–4 weeks with no GI distress, you have a choice: hold at 40 units indefinitely (this dose alone produced meaningful effects in the Phase 2 4 mg-arm, when scaled) or begin the Phase 2 monthly titration to higher target doses.
The Phase 2 trial used 2 mg weekly as the starting dose for all dose arms. That's the "reference" induction dose — below it, no published data exists; above it, GI side effects spike sharply.
The 20 / 30 / 40 ramp is slower than the trial protocol. Instead of jumping into 2 mg (40 units) on week 1, you spend two weeks below it — at 1 mg, then 1.5 mg — letting the gut adapt to GP-1 receptor signaling before reaching the trial-validated starting point.
Why this works:
Phase 2 trial subjects had support infrastructure — clinical staff, anti-emetic protocols, weekly check-ins. Most independent research protocols don't, which justifies a gentler ramp.
These unit numbers depend on a specific reconstitution. If you change the bac water volume, you change the per-unit dose. The standard:
So: - 20 units × 0.05 mg = 1.0 mg - 30 units × 0.05 mg = 1.5 mg - 40 units × 0.05 mg = 2.0 mg
For the 20mg vial, double the bac water to 4 mL to keep the same 5 mg/mL concentration and the same unit math. That vial gives you ~10 weekly doses at 2 mg, which covers weeks 3–12 of a beginner protocol with comfortable buffer.
If you reconstitute at a different concentration (e.g., 10 mg/mL with 1 mL bac water), the unit math changes. The protocol numbers above only work at 5 mg/mL. See our reconstitution guide for general technique.
All Phase 2 reta trials used once-weekly subcutaneous administration. The peptide's albumin-binding fatty acid chain gives it a half-life of approximately 6 days, so weekly dosing produces near-steady-state exposure.
Practical pointers:
The protocol is gentle, but individual responses vary. Hold at the current dose (don't escalate next week) if:
Step down (reduce to the previous week's dose) if:
There's no penalty for taking longer to ramp. The endpoint is the same; the timeline is different.
For longer protocols, our peptide storage guide covers freeze-thaw cycles and stability extension.
The 20/30/40 ramp is for researchers new to GP-class compounds. It's not the right protocol if:
A 12-week beginner protocol uses: - 2 vials of reta 10mg (or 1 vial of reta 20mg) - 1 vial of bacteriostatic water 10mL - A few standard U-100 insulin syringes
The Metabolic Stack (Reta 20mg + Tesamorelin 10mg) is the cleanest entry point for researchers also studying body composition rather than body weight alone — see our reta vs tesamorelin discussion for the stacking rationale.
Use code WELCOME at checkout for 35% off reta 10mg, reta 20mg, bacteriostatic water, and the Metabolic Stack. Every vial ships with a batch-matched COA showing 99%+ HPLC purity, US-based handling, and same-batch consistency for multi-vial protocols.
